Beutler

October 27 - November 1, 2010 • Sheraton Seattle Hotel • Seattle, WA

Meeting Summary

Vaccines have been described as one of the most cost effective public health tools in history. Despite their success, most vaccines have been made empirically, and we understand little about the mechanisms by which they stimulate protective immunity. Indeed, immunologists and vaccinologists have until recently, occupied parallel universes! Understanding the immunology of vaccines and infections is of paramount importance in the rational design of future vaccines against pandemics such as HIV, malaria and tuberculosis, and against emerging infections such as swine influenza and dengue. Recent advances in immunology, human genetics, and systems biology are beginning to offer a deeper understanding of the molecular and cellular mechanisms of host immunity, and the pivotal role played by the innate immune system in shaping the adaptive immune response. Such insights are enhancing our understanding of the correlates of protective immunities, of the early innate immune signatures of vaccine efficacy, and guiding the rational design of novel and safer vaccines and adjuvants. Immunological Mechanisms of Vaccination will bring together an interdisciplinary group of scientists in immunology, virology, systems biology and vaccinology, to address critical advances in these fields and their implications for vaccinology. • The importance of the innate immune system in sensing vaccines and shaping the vaccine immunity • Modulating immune memory to vaccines with innate immunity • Novel approaches to understanding host-pathogen interactions • Deconstructing the mode of action of successful vaccines, vectors adjuvants with innate immunity • Constructing new vaccines and adjuvants with innate immunity • Application of systems biology to predicting vaccine efficacy, and discovering novel immune mechanisms of protection

One naive T cell, multiple fates in CD8+ T cell differentiation

Ton Schumacher's group has developed a system that allows for thousands of unique "barcodes" to be imprinted to a naive CD8+ T cell population. This technique allows them to ask if a single naive precursor develops into diverse populations of effector and memory T cells, or if each individual cell is programmed to a specific fate. Highly recommended reading...

http://jem.rupress.org/content/early/2010/05/12/jem.20091175

ABSTRACT: The mechanism by which the immune system produces effector and memory T cells is largely unclear. To allow a large-scale assessment of the development of single naive T cells into different subsets, we have developed a technology that introduces unique genetic tags (barcodes) into naive T cells. By comparing the barcodes present in antigen-specific effector and memory T cell populations in systemic and local infection models, at different anatomical sites, and for TCR–pMHC interactions of different avidities, we demonstrate that under all conditions tested, individual naive T cells yield both effector and memory CD8+ T cell progeny. This indicates that effector and memory fate decisions are not determined by the nature of the priming antigen-presenting cell or the time of T cell priming. Instead, for both low and high avidity T cells, individual naive T cells have multiple fates and can differentiate into effector and memory T cell subsets.

from the Portland Business Journal - Drug manufacturer Genentech is weeks away from starting production of pharmaceuticals at its $400 million Hillsboro campus.

The San Francisco-based company already employs 250 across its local warehouse and packaging lines. As it ramps up biotech manufacturing over the next four years, it expects to add 50 more employees.

On Tuesday Genentech opened its doors to Gov. Ted Kulongoski, the media and other officials to highlight its rapid growth.

Kulongoski aggressively courted the company, and backed a $4.8 million investment in Genentech's move to Oregon. That investment includes a $2 million loan to fund worker training that Genentech will not have to repay because it has already exceeded its job creation target of 200 employees.

"What Genentech brings to us is the opportunity for a seat in a biotechnology sector that will be here for years and years and year," Kulongoski said.

Genentech officials credited Kulongoski and other government officials for their decision to pick Oregon, and said they were also drawn by the state's business climate. Genentech vetted a number of states before ultimately choosing Oregon.
"The tax climate here was very favorable, certainly, when compared to California," said Barry Starkman, manager of the Hillsboro plant.

The company announced its plans to relocate some of its San Francisco manufacturing and distribution to Oregon in 2006. It bought its 75-acre Hillsboro campus that year, and by 2008 had opened its distribution warehouse. It later added a finishing line, where workers oversee an array of machines that lables to drugs made elsewhere, then package the labled drugs for distribution.

Later this month, it will begin manufacturing its first batches of pharmaceuticals, as part of the longer and more thorough process of winning U.S. Food and Drug Administration approval to make and sell drugs commercially.

The company hopes to win approval for its first production line by the first quarter of 2011, and to be making Avastin, Rituxan and Hereptin for domestic and international use by the end of 2013.
